Our research is aimed to understand plant cell responses and acclimation and adaptation strategies to environmental stresses. Furthermore we aim to elucidate molecular mechanisms, which are involved in assembly, function, maintenance and regulation of the photosynthetic machinery in oxygenic chloroplast photosynthesis. For this purpose, we are mainly using the green alga Chlamydomonas reinhardtii as a model system and combining molecular techniques like reverse genetics and proteomics to elucidate these processes. Moreover we are also interested in exploring the biotechnological potential of microalgae. Proteomics has been a key technique for the laboratory. In the last years we established quantitative proteomics techniques using isotopic metabolic labelling. Hereby we are employing differential 14N/15N metabolic labeling as well as stable isotopic labeling with amino acids in cell culture (SILAC) to monitor protein dynamics and multi-protein complex compositions. Moreover we are engaged in the development of mass spectrometric data analysis tools. Accordingly proteomics is used as a key technology to investigate research topics A-E (see below).
