Chembion project description:
Hyperpolarization-activated cyclic nucleotide-gated cation (HCN) channels play a key role in the regulation of cardiac and neuronal rhythmicity. An HCN subunit is comprised of six α-helices (S1–S6) and a loop domain between the S5 and S6 helix that forms the ion selectivity filter and the positively charged S4 helix functioning as a voltage sensor. The cyclic nucleotide-binding domain (CNBD) is connected to the channel core (S6 helix) via the C-linker. cAMP binding to CNBD leads to induction of a rightward shift in the voltage-dependence of activation. The molecular structure of the CNBD in complex with cAMP was resolved for all four channel subtypes.
